Study of Reticular Celis By the Aid of A Metallophic Technigue

Öz

Recentiy identified Iymphoid reticulum celi types are of special interest because of their functional role in the immune system, A plastic embedding metallophilic technigue (ZİO — zinc iodide-osmium tetroxide) identifying different types of reticulum cells in the normal cytoarchitecture of the Iymphoid organs İs presented. This technigue enables revealing the morphological relationship between the Iymphocytes and specific reticulum cells in both light and electron microscopic levels and also differentiates follicular dendritic cells (FDC), tingible body macrophages (TBM) and some other macrophage groups. Our findings clearly reveal the fine structure of FDCs and TBMSs. In addition there are some clues that this technigue can be applied on the same section together with enzyme or immuno-histochemical methods. We hope
that the presented technigğue will be used in many laboratories, being relatively easier and cheaper, in ontogenetic and histopathological studies,

Retiküler Hücrelerin Metalofilik Bir Yöntemle (Çıo) İncelemesi: Işık Ve Elektron Mikroskobu Düzeyinde Çalışma

Öz

Yeni tanımlanan lenfoid retiküler hücre tipleri bağışıklık sistemindeki işlevsel rolleri hedeniyle büyük önem kazanmışlardır. Lönfoid dokuların normal sitoarkitektürü içinde retiküler hücre tiplerini , ayırt eden bir metalofilik (ÇİO — Çinko iyodit-osmiyum tetroksit) “plastik gömme tekniği sunulmuştur. “Bu teknik ışık ve elektron mikroskobu düzeyinde lenfositler ve retiküler hücreler arasındaki yapısal ilişkiyi açığa çıkarmakta; folliküler dendritik hücreler (FD), tingible body makrofajları. (TBM) ve diğer makrofaj gruplarının tanımlanmasını sağlamaktadır. Bulgularımız FDH ve TBM'lerin ince yapı özelliklerini ortaya koymaktadır. Ek olarak retiküler hücrelerin tanımlan- - ması amacıyla kullanılan enzim histokimyası ve immunohiştokimyasal yöntemlerin aynı kesitler üzerinde birlikte uygulanabileçeğine dair ipuçları vardır. Sunulan teknik bu özellikleriyle ontogenetik ve histopatolojik çalışmalarda yaygın olarak em cl olup göreceli olarak kolay ve ucuz bir yöntemdir.
